With the rapid development of the biopharmaceutical industry, the scalability of the fermentation system has become a key factor restricting the scale-up of the industry. This paper discusses the challenges and opportunities of fermentation system scalability in the biopharmaceutical industry from the perspectives of innovation and practice, and puts forward corresponding strategies and suggestions, with a view to providing references for the scale-up of biopharmaceutical enterprises.



Biopharmaceuticals, as a high-tech industry, have been developing rapidly worldwide. Fermentation technology, as the core process of biopharmaceuticals, and its scalability directly affects the production efficiency and cost control of products. However, the scale-up of fermentation systems is not easy and faces many challenges. The aim of this paper is to explore how to improve the scalability of fermentation systems through innovation and practice to promote the healthy development of the biopharmaceutical industry.

Challenges to fermentation system scalability

1. Bioreactor scale-up effect: During the scale-up process of a fermentation system, the parameters such as the flow field, temperature field, and the distribution of dissolved oxygen in the bioreactor change, which may lead to the differences in the growth of the cells and metabolites.

2. Difficulty in optimization of process parameters: With the expansion of fermentation scale, the original laboratory process parameters may no longer be applicable and need to be re-optimized and adjusted.

3. Production cost control: large-scale production requires more investment in equipment and operating costs, how to maintain product quality while reducing costs has become a major problem.

4. Production Stability: How to maintain the stability of the production process and reduce the batch-to-batch variation during large-scale fermentation is an important challenge for the scalability of the fermentation system.

Innovation and practice of fermentation system scalability

1. Innovation of bioreactor design

(1) Novel stirrer paddle design: Computational Fluid Dynamics (CFD) technology is used to design stirring paddles that are more suitable for large-scale fermentation stirring paddles to improve mixing efficiency and reduce shear force.

(2) Multi-parameter control system: Integrate the multi-parameter control system of temperature, pH, dissolved oxygen, rotational speed, etc., to realize the precise control of fermentation process.

2. Optimization practice of fermentation process

(1) Systematic research on process parameters: establish a database of key process parameters of the fermentation process through experiments and data analysis to provide reference for large-scale production.

(2) Online monitoring and control: using sensors and data analysis technology, real-time monitoring of the fermentation process to achieve dynamic optimization of process parameters.

3. Fermentation process simulation and scale-up strategy

(1) Process simulation: using computer simulation technology to predict the behavior of the fermentation process at different scales, and to provide a theoretical basis for actual scale-up.

(2) Phased scale-up: gradually scaling up from laboratory scale to production scale, with each phase focusing on optimization and validation of process parameters.

Opportunities for fermentation system scalability

1. Improve production efficiency: The production efficiency of biopharmaceutical companies can be significantly improved and time-to-market can be shortened through the enhancement of fermentation system scalability.
